7 Medications That May Affect Memory and Dementia Risk

The claim that certain medicines “cause severe dementia and profound brain damage” is too strong. Research has found associations between long-term exposure to medications with strong anticholinergic effects and increased dementia risk, particularly in older adults, but these studies do not prove that the medications directly cause dementia or permanent brain damage. Examples of medications/classes worth reviewing with a doctor include:

  1. Certain tricyclic antidepressants — such as amitriptyline and some related drugs.

  2. Some bladder medications — particularly antimuscarinics such as oxybutynin.

  3. First-generation antihistamines — some older allergy/sleep medicines have strong anticholinergic effects.

  4. Certain antipsychotic medications — some have substantial anticholinergic activity.

  5. Certain antiparkinsonian medications — particularly older anticholinergic drugs.

  6. Some antiepileptic medications — certain drugs in this category have anticholinergic properties.

  7. Certain medications for dizziness or nausea — some antivertigo/antiemetic drugs can have anticholinergic effects.

A large observational study of adults 55+ found higher cumulative exposure to several strong anticholinergic classes was associated with increased dementia risk. However, the researchers explicitly noted that the observational design could not establish causality. JAMA Network

Don’t stop your medication suddenly

If you take any of these medicines, don’t stop them on your own. A doctor or pharmacist can review your complete medication list, determine whether a drug has anticholinergic activity, and consider safer alternatives when appropriate.

Also, short-term confusion or memory problems caused by some medications can be different from dementia and may improve after the medication is changed under medical supervision. JAMA Network
